Prismatic Jelly will always drop from killing Prismatic Slimes. They are only obtainable during one of the Wizard's Special Orders.

Multiple jellies may be obtained, by finding more prismatic slimes before completion of the quest or by having a Burglar's Ring. However only one is required for the Wizard, and as a quest item, any others will disappear from the player's inventory once the quest is complete.

While the quest is active, ca. 1 of 83 Slimes is a Prismatic Slime, that's a likelihood of about 1.2% that a Prismatic Slime replaces a normal Slime.
